Priyabandhavi unfolds in a rural setting and follows Radha (played by Saradha), a sincere and compassionate schoolteacher devoted to her work and her aging father. Venu (played by Ranganath), a respected young doctor, gradually falls in love with her quiet strength and integrity. With the approval of their families, their marriage is arranged, but the alliance collapses dramatically when a shocking claim surfaces that Radha already has a six-year-old daughter. Humiliated and heartbroken, Radha becomes the target of village gossip, while Venu, unable to uncover the truth behind the allegation, reluctantly agrees to marry another woman, Madhavi (played by Roopa). The scandal devastates Radha's father, who loses his sanity, forcing Radha to leave the village and rebuild her life elsewhere.
Years later, Radha works in a convent school where she raises the young girl Vani as her own while caring for her ailing father. Fate brings Venu back into her life when his son studies in the same school. Through a series of revelations, Venu learns the truth: Vani is not Radha's daughter but the child of Geetha, Radha's close friend and Venu's former lover from his youth, whom Radha had selflessly promised to protect. The discovery shatters Venu, who realizes that the misunderstanding that destroyed his relationship with Radha had concealed the existence of his own child.
As Venu begins to reconnect with his daughter Vani, his wife Madhavi senses the buried secrets surrounding her husband's past. When the truth about Vani's parentage finally comes to light, the child is devastated to learn that Radha is not her real mother. Overwhelmed by the feeling of abandonment, Vani attempts to end her life. In a desperate race against time, Radha saves the girl from certain death but sacrifices her own life in the process, leaving the child in the care of her real father Venu and his compassionate wife Madhavi, while her own legacy remains one of boundless love and sacrifice.
